Morphological and cytochemical characteristics of neutrophil in the marbled rockfish,Sebastiscus marmoratus were examined by light microscopy. Two types of granules, chromophobicgranule(βG)and basophilic granule(γG)were observed in the neutrophil. Multiple Romanowsky-type stain valuation revealed that γG was stained light blue with May-Grünwald(MG)and reddish purple with MG-Giemsa, but not stained with Giemsa. These findings indicate that the content(s) of γG is both orthomethylenophilic and metaazurophilic, and the content(s) of γG dissolves in methanol(fixative before Giemsa stain)but is fixed with methylene blue eosinate in methanol(MG solution). The βG was negative with Romanowsky-stain but positive with peroxidase stain.
