Two types of extraordinary chromophobic granules (type 1, eoG-1^{Cd}; type 2, eoG-2^{Cd}) were observed in the neutrophils of red seabream Pagrus major infected with Clavellotis dilatata (parasitic copepod). The eoG-1^{Cd} showed chromophobic, simple morphology (without stratified structure), peroxidase (PO) positive, Sudan black B (SBB) negative and lacked lysozomal enzymes. The eoG-2^{Cd} was stratified granule with two-layer structure [inner eosinophilic layer (L0) and outer chromophobic layer (L1)] and SBB negative. Alpha-naphtyl butyrate esterase and PO were detected in L0 and L1, respectively. Spot formation, a curious phenomenon appeared in PO-stained type 2 ordinary chromophobic granule (oG-2; positive L1 and negative L0) from non-infected fish, was not observed in the infected fish. Acid phosphatase (AcP), β-glucuronidase (β-Glu), α-naphtyl acetate esterase (α-NAE) and naphthol AS-D chloroacetate exterase (CAE) were detected in L0 of eoG-2^{Cd} as small central positive particles. These findings indicate that the L0 of eoG-2^{Cd} consists of L0-0 (inner part in L0; AcP, β-Glu, α-NAE and CAE positive) and L0-1 (outer part of L0-0).
